Description

La diversidad microbiana es un recurso poco estudiado en México. En zonas como Sinaloa en donde la agricultura es un importante motor socioeconómico, la mayor parte de la superficie es empleada para cultivos agrícolas. Este es el caso del municipio de Guasave, en donde la zona de preservación ecológica de centro de población La Uva constituye el único relicto de vegetación original preservada. Es de vital importancia iniciar esfuerzos a nivel local y nacional para conocer y conservar la diversidad microbiana presente en los suelos de esta región y del país en general. Una de las dificultades en el estudio de la diversidad microbiana es que se calcula que sólo se pueden cultivar en el laboratorio del 1 al 5% de la diversidad total de microorganismos. Por lo tanto, la utilización de técnicas moleculares de identificación en las que no se involucre el aislamiento de microorganismos resulta una estrategia que permite describir de manera más real la diversidad de los microorganismos presentes en un ecosistema tan diverso como lo es el suelo. En nuestro grupo iniciamos un primer esfuerzo a partir del 2006 a través de la generación de un banco de germoplasma vivo de microorganismos de suelo asociados a la rizosfera de plantas de tomate en un campo agrícola de la región, el cual consiste de cerca de 750 aislados de microorganismos preservados en ultracongelación a -70°C. Del mismo modo se generó información de la diversidad microbiana total a través del aislamiento de ADN genómico de suelo, y la posterior generación y secuenciación de bibliotecas de ADNr. Mediante esta estrategia se identificaron 500 fragmentos de ADN de origen procariote y 500 de origen eucariote. En el presente proyecto primeramente se propone concluir la identificación de los microorganismos existentes en el banco de tomate para posteriormente utilizar la misma estrategia de generación de un banco de microorganismos vivos y un banco de DNA de suelo a partir de muestras de rizosfera de Datura stramonium, la cual es una planta solanácea silvestre común en la zona de reserva. Posteriormente esta información será utilizada para realizar estudios comparativos entre tipo de organismos presentes en la rizosfera de solanáceas en zonas de cultivo, tal como el tomate, y solanáceas en zonas preservadas. El objetivo del trabajo en la zona de reserva incluirá el análisis de la diversidad de microorganismos asociados a la rizosfera de D. stramonium a través de la secuenciación masiva de 1,000 fragmentos de ADN ribosomal de una biblioteca generada a partir de ADN genómico de suelo y la generación de un banco de germoplasma vivo de 750 especimenes criopreservados a -70°C. Finalmente se propone elaborar una base de datos compatible con el sistema Biótica con los datos recabados para cada espécimen del banco, el cual incluirá los datos de colecta, georeferencias, características físicas y químicas de los suelos, fotografías que muestren la morfología colonial y microscópica de cada aislado, los datos del procedimiento de identificación molecular y análisis de secuencias. Además se pondrán a disposición los datos de las listas de las especies identificadas en los dos bancos de germoplasma vivos y en los bancos generados a partir de los datos de secuenciación masiva de ADN ribosomal tanto de los suelos de la zona de cultivo como los de la zona de reserva.
