The InBIO Barcoding Initiative Database: A taxonomic revision of the Western Palaearctic genus Cacochroa Heinemann, 1870 (Lepidoptera, Depressariidae)

The dataset contains 11 records of moth specimens (Lepidoptera, Depressariidae) collected from 1988 to 2016 in seven South European and North African countries. The study of these specimens resulted in the description of a new genus, Rosetea Corley & Ferreira, 2019. The new genus contains two species previously placed in the genus Cacochroa Heinemann, 1870, Rosetea corfuella (Lvovsky, 2000) and R. rosetella (Corley, 2018), but also a new species, R. sara Corley & Ferreira, 2019, described based on differences found in the male and female genitalia and on DNA barcodes in Corley et al. (2019). Captured specimens were identified to species level and preserved in 96% ethanol, with one exception kept in dry storage. A tissue sample, one leg, was collected from each individual, from which DNA was extracted. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA barcode sequences were deposited in BOLD (Barcode of Life Data System) online database. The InBIO Barcoding Initiative (IBI) aims to develop a reference collection of DNA barcoding sequences primarily covering Portuguese invertebrate taxa providing a tool in ecological studies.

The DNA barcoded specimen morphological identification was contrasted with the results of the Blast of the newly generated DNA barcode in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).Method steps

- The specimen was morphologically identified using a binocular stereoscopic microscope and resorting to available literature (Corley, 2018; Corley. & Ferreira 2019; Heinemann, 1870; Herrich-Schäffer, [1853−1855] 1847−1855; Lvovsky, 1981 and 2000; Rebel, 1901).
- DNA barcodes were sequenced from all specimens. For this, one leg was removed from each individual from which a 658 bp COI DNA barcode fragment was extracted, amplified and sequenced.
- The obtained sequences were submitted to BOLD database and the specimens’ morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcode in BOLD Identification Engine.
- Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
Taxonomic Coverages
This dataset is composed of data relating to 11 Lepidoptera specimens. All specimens were determined to species level.
Five species belonging to two genera, Cacochroa Heinemann, 1870 and Rosetea Corley & Ferreira 2019, are represented in the dataset. Both genera belong to the family Depressariidae.
