The dataset contains one record of the species Borkhausenia crimnodes (Lepidoptera, Oecophoridae) collected in continental Portugal in 2014. This species, a southern hemisphere moth, had not been recorded in Europe before. The specimen was detected and captured by direct search of the environment and preserved in 96% ethanol. A tissue sample, one leg, was removed to be used for DNA extraction. Sequencing of the 658 bp COI DNA barcode was conducted within the InBIO Barcoding Initiative (IBI) and the DNA sequence was submitted to BOLD (Barcode of Life Data System) and GenBank databases. The preserved specimen and DNA extract are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).Purpose
The InBIO Barcoding Initiative (IBI) aims to develop a reference collection of DNA barcoding sequences primarily covering Portuguese invertebrate taxa providing a tool in ecological studies.
Sampling Description
Study Extent
A single specimen was collected in Louriçal, in Continental Portugal, in 2014.Sampling
The Borkhausenia crimnodes specimen was captured in continental Portugal in 2014, by direct search of the environment. The captured specimen was preserved in 96% ethanol and stored at the InBIO Barcoding Initiative reference collection for downstream molecular analysis. The collected specimen was morphologically identified.Quality Control
The DNA barcoded specimen morphological identification was contrasted with the results of the Blast of the newly generated DNA barcode in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).Method steps
- The Borkhausenia crimnodes specimen was captured in a single locality of continental Portugal, during 2014.
- The specimen was captured by direct search of the environment. The captured specimen was preserved in 96% ethanol.
- The specimen was morphologically identified using a binocular stereoscopic microscope and resorting to available literature (Meyrick, 1912).
- A DNA barcode was sequenced from a tissue sample, one leg, that was removed. DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced.
- The obtained sequence was submitted to BOLD and GenBank databases and the specimen morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcode in BOLD Identification Engine.
- Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
