The community composition of suspended protists was investigated at 3 stations on the East Greenland continental shelf as part of the EU project ECOTIP. Water was sampled with a 1.7 L Nansen Water bottle at the stations LCC1 (13 May 2022), LCC2 (14 May 2022), and LCC3 (17 May 2022) at 5, 15, 35 m. At station LCC2, a sample was also collected at 90 m. Water samples were collected through a hole in the land-fast sea ice at station LCC1 and LCC2 and from a ship at station LCC3 (station located in a polynya outside Scoresby Sound). The collected water was gently transferred into a carboy, which was stored cool (not freezing) and in darkness until handling within few hours. From each carboy, a subsample of 100 mL was transferred into a glass bottle, preserved with 1 mL acidic Lugol’s solution for later microscopic analysis. Study Extent
At 3 sampling stations (LCC1, LCC2, LCC3) on the East Greenland continental shelf, water was collected in repeated casts of a Nansen water bottle at 5, 15, and 35 m. In addition, water was collected at 90 m at station LCC2. While the water was collected through a hole in the land-fast sea ice at station LCC1 and LCC2, the water was collected at LCC3 from a ship (located in a polynya outside Scoresby Sound).Sampling
At each station, the water was gently transferred from the 1.7 L Nansen water bottle to a carboy (using a silicon hose). The carboy was stored cool and in darkness until handling aboard the ship Le Commandant Charcot. There, a subsample of 100 mL was transferred from the carboy to a glass bottle and preserved with 1 mL acidic Lugol’s solution. The sample was stored cool (+4 C) and in darkness until further analysis ashore.Method steps
- The microscopic analysis was later conducted at the Institute of Oceanology of the Polish Academy of Sciences (IO PAN) in Sopot, Poland and the phytoplankton taxa were identified to the lowest possible taxonomic level.
