The dataset contains five records of moth specimens of the genus Isotrias collected from 2012 to 2016 in continental Portugal. The study of these specimens resulted in the discovery of sexual dimorphism in the species Isotrias penedana Trematerra, 2013, and in the description of the previously unknown female of the species in Corley and Ferreira (2017). Specimens were detected and captured by direct search of the environment during the day, with adults’ hand-netted, and by night using mercury-vapor lamps to attract the insects. Captured specimens were identified to species level and kept in dry storage. A tissue sample, a leg, was collected from each individual, from which DNA was extracted. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA barcode sequences were deposited in BOLD (Barcode of Life Data System) and GenBank online databases. DNA extracts are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).Purpose
The InBIO Barcoding Initiative (IBI) aims to develop a reference collection of DNA barcoding sequences primarily covering Portuguese invertebrate taxa providing a tool in ecological studies.

The DNA barcoded specimen morphological identification was contrasted with the results of the Blast of the newly generated DNA barcode in BOLD Identification Engine. Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).Method steps

- The captured specimens were preserved in dry storage.
- The specimen was morphologically identified using a binocular stereoscopic microscope and resorting to available literature (Corley & Ferreira, 2017, Razowski, 2002; Trematerra, 1991, 2013).
- DNA barcodes were sequenced from all specimens. For this, one leg was removed from each individual from which a 658 bp COI DNA barcode fragment was extracted, amplified and sequenced.
- The obtained sequences were submitted to BOLD and GenBank databases and the specimens’ morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcode in BOLD Identification Engine.
- Prior submission to GBIF, data was checked for errors and inconsistencies with OpenRefine 3.3 (http://openrefine.org/).
Taxonomic Coverages
This dataset is composed of data relating to 5 Lepidoptera specimens. All specimens were determined to species level. All specimens belong to two species of the genus Isotrias Meyrick, 1895 and the family Tortricidae.
