Bartonella spp. was screened in 634 phlebotomine sand flies specimens, belonging to 44 different sand fly species, sampled between 2017-2021 in North and Northeastern Brazil. Detection by quantitative real-time polymerase chain reaction (qPCR) targeting the 16S-23S ITS intergenic region, found 8.7% (55/634) of the specimens positive for Bartonella. Brazilian sand flies do harbour Bartonella and might be involved the epidemiological cycle of these agents.

Specimens were captured using CDC or Shannon traps in ecological reserves and parks from Brazil: preserved forest areas in the cities of Xapuri and Rio Branco (Acre); Murici Ecological Station (Alagoas); Pau Brasil National Park (Bahia); Ubajara National Park (Ceará); Tapajós National Forest (Pará); Dois Irmãos State Park (Pernambuco); Viruá National Park (Roraima).Sampling
In the preserved forest areas in the cities of Xapuri and Rio Branco (Acre) sampling was carried out monthly. In other sites, sampling was a one week opportunistic field work.Quality Control

- Sand flies were collected using wither CDC light traps or Shannon traps to assess anthropophily. DNA was extracted from dissected sand flies, only thorax and abdomen as heads and three last abdominal segments were used for morphological identification using the TRIzolTM (Invitrogen®, Thermo Fisher Scientific, https://www.thermofisher.com/), following the manufacturer instructions. The presence of potential PCR inhibitors was assessed using a conventional PCR based on the cytochrome c oxidase subunit-1 (cox-1) endogenous gene for invertebrates.
